Alter macrophage adhesion and modulate their response on hydrophobically modified hydrogels

چکیده
Adhesion of macrophages to material substrates plays an essential role in many biological behaviors and biomedical procedures, including wound healing, cancer treatment biomaterial implantation. Therefore, altering the adhesion using design may help control their function. In this work, we developed a straightforward method render hydrogel surface more hydrophobic demonstrated that on hydrogels was enhanced these surfaces. Compared unmodified methacrylated gellan gum (MGG) hydrogel, water contact angle increased swelling ratio decreased for modified MGG hydrogels. The degree hydrophobicity controlled by attaching branches different lengths MGG, while modification did not change compressive modulus. This ensured observed cell responses were mainly due mechanical property difference among Cell (RAW 264.7 macrophages) proliferation spreading with increasing hydrophobicity. Protein adsorption surfaces showed same trend. Quantification probe as substrate increased. suggests be promoted increase could enhance protein adsorption. addition, tumor necrosis factor-? (TNF-?) secretion lipopolysaccharide (LPS) activated M1 interleukin-4 (IL-4) M2 also altered TNF-? production, marker pro-inflammatory macrophages, These results suggest can used modulate macrophage responses.

Drug diffusion in hydrophobically modified N,N-dimethylacrylamide hydrogels
The use of hydrophobically modified hydrogels for drug release was investigated. Copolymers of N,N-dimethylacrylamide and 2-(N-ethylperfluorooctanesulfonamido) ethyl acrylate (FOSA) were prepared by free-radical polymerization. The drug release rates, dynamic swelling behavior, and pH sensitivities of copolymers ranging in composition from 0 to 30 mol% FOSA were studied. Pheniramine maleate, an...
